Daratumumab + bortezomib + dexamethasone is a three-drug combination regimen used primarily for the treatment of multiple myeloma, particularly in patients who have received at least one prior therapy. Daratumumab is a monoclonal antibody that targets CD38, a protein highly expressed on myeloma cells, leading to direct cell death and immune-mediated cytotoxicity. Bortezomib is a small molecule proteasome inhibitor that disrupts protein degradation within cancer cells, resulting in apoptosis. Dexamethasone is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressive properties; it also enhances the efficacy of the other agents in this combination. This regimen has demonstrated significant improvement in progression-free survival compared to bortezomib and dexamethasone alone[3][4][5][9].
