Daratumumab + lenalidomide is a combination therapy used primarily for the treatment of multiple myeloma, a type of blood cancer. Daratumumab is a monoclonal antibody that targets CD38, a protein highly expressed on the surface of myeloma cells. By binding to CD38, daratumumab induces cell death through several mechanisms including antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), antibody-dependent cellular phagocytosis (ADCP), and direct induction of apoptosis via Fc receptor-mediated crosslinking. Lenalidomide is an immunomodulatory agent with antiangiogenic properties; it enhances immune effector cell function and inhibits tumor growth by preventing new blood vessel formation in tumors. The combination has demonstrated significant efficacy in prolonging progression-free survival in patients with relapsed or refractory multiple myeloma[1][2][5][6].
