Daratumumab + teclistamab is a combination of two monoclonal antibodies used in clinical trials for the treatment of relapsed or refractory multiple myeloma. **Daratumumab** is a monoclonal antibody that targets CD38, a surface protein highly expressed on myeloma cells, leading to cell death through mechanisms including immune-mediated cytotoxicity, complement activation, and direct apoptosis[8]. **Teclistamab** is a bispecific monoclonal antibody that binds both BCMA (B-cell maturation antigen) on myeloma cells and CD3 on T cells, redirecting T cells to kill BCMA-positive myeloma cells[2][6]. The combination provides a dual immunotherapy approach, leveraging both direct tumor cell targeting and immune system redirection, and aims to improve outcomes in heavily pretreated and relapsed/refractory multiple myeloma. This combination is under evaluation in phase 1b/2/3 clinical trials and has shown promising efficacy and manageable safety profiles in early studies[1][9][10].
