Overview

This combination regimen consists of the non-steroidal androgen receptor (AR) antagonist darolutamide, the gonadotropin-releasing hormone (GnRH) agonist goserelin, and the human epidermal growth factor receptor 2 (HER2)-targeted monoclonal antibodies pertuzumab and trastuzumab. It is being evaluated in a Phase 2 umbrella trial (NCT05933408) led by Peking Union Medical College for the treatment of salivary gland carcinoma (SGC). The regimen is specifically designed for patients whose tumors exhibit dual expression of AR and HER2, a molecular profile often found in salivary duct carcinoma. By simultaneously inhibiting the AR signaling pathway (via darolutamide and goserelin) and the HER2 pathway (via pertuzumab and trastuzumab), the therapy aims to overcome potential resistance mechanisms and improve clinical outcomes in this biomarker-selected population.
