darotropium + salmeterol

GSK233705B + salmeterol is a dual bronchodilator combination therapy developed by GSK for the treatment of chronic obstructive pulmonary disease (COPD). The combination consists of darotropium (GSK233705), an inhaled long-acting muscarinic antagonist (LAMA), and salmeterol, a long-acting beta-2 adrenergic receptor agonist (LABA). Darotropium acts as a high-affinity pan-active muscarinic receptor antagonist, inhibiting the action of acetylcholine to prevent bronchoconstriction, while salmeterol directly relaxes bronchial smooth muscle by stimulating beta-2 adrenoceptors. This combination was investigated in Phase II clinical trials (NCT00422604) to assess its efficacy in providing superior bronchodilation compared to monotherapy. However, development of this specific twice-daily combination was not progressed further as GSK prioritized the development of once-daily alternatives such as the combination of vilanterol and umeclidinium.
