A fixed-dose combination antiretroviral drug composed of four small molecule agents: **darunavir**, a protease inhibitor; **cobicistat**, a pharmacokinetic enhancer (booster) via CYP3A inhibition; **emtricitabine**, a nucleoside reverse transcriptase inhibitor (NRTI); and **tenofovir disoproxil fumarate**, a nucleotide reverse transcriptase inhibitor (NtRTI). This combination is used for the treatment of HIV-1 infection in adults and pediatric patients. Darunavir inhibits the cleavage of HIV polyproteins, preventing viral replication. Cobicistat increases the plasma concentration of darunavir by inhibiting its metabolism via CYP3A. Emtricitabine and tenofovir disoproxil fumarate block HIV reverse transcriptase, essential for viral RNA to DNA conversion. The combination helps suppress viral load and delay disease progression but is not a cure for HIV/AIDS.
