Overview

Dasatinib + lenalidomide is a combination of two small molecule drugs used primarily in the treatment of hematologic malignancies, including multiple myeloma and Philadelphia chromosome-positive chronic myeloid leukemia. Dasatinib is a tyrosine kinase inhibitor that targets BCR-ABL, SRC family kinases (SRC, LCK, YES, FYN), c-KIT, EPHA2, and PDGFRβ. It inhibits both the active and inactive conformations of the ABL kinase domain to block proliferation and survival signals in cancerous hematopoietic cells[4]. Lenalidomide is an immunomodulatory agent that alters cytokine production, enhances natural killer cell-mediated cytotoxicity, and modulates substrate specificity of the CRL4 CRBN E3 ubiquitin ligase complex. This leads to degradation of key transcription factors (IKZF1/IKZF3) essential for malignant B-cell survival[5][6]. The combination has demonstrated feasibility and preliminary clinical benefit in heavily pre-treated populations with manageable toxicity profiles[2].
