This is a combination regimen of three pharmaceutical agents—dasatinib, mitoxantrone, and cytarabine—used primarily in the treatment of certain leukemias. Dasatinib is a small molecule tyrosine kinase inhibitor that targets BCR-ABL fusion protein and other kinases such as SRC family kinases and c-KIT proto-oncogene receptor tyrosine kinase. It is approved for Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) and chronic myeloid leukemia (CML)[3]. Mitoxantrone is an anthracenedione antineoplastic agent that intercalates into DNA and inhibits topoisomerase II alpha activity. Cytarabine is an antimetabolite chemotherapeutic agent that inhibits DNA synthesis by incorporating into DNA during the S phase of the cell cycle[5][6]. This combination has been explored in regimens for mixed phenotype acute leukemia with Philadelphia chromosome positivity (Ph+ MPAL), aiming to leverage both myeloid- and lymphoid-targeted chemotherapy alongside targeted inhibition of BCR-ABL fusion protein[1][2].
