Overview

Dasatinib + vincristine + prednisone is a combination regimen used primarily as maintenance therapy for Philadelphia chromosome–positive (Ph+) acute lymphoblastic leukemia (ALL). Dasatinib is an oral, multitargeted tyrosine kinase inhibitor that targets BCR-ABL1 and SRC family kinases, effective against many imatinib-resistant mutations. Vincristine is a vinca alkaloid that inhibits microtubule formation in mitosis, leading to cell cycle arrest. Prednisone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties, inducing apoptosis in certain cancer cells. This combination aims to suppress leukemic cell proliferation through complementary mechanisms—targeted inhibition of oncogenic kinases (dasatinib), disruption of mitosis (vincristine), and induction of apoptosis via steroid signaling (prednisone). The regimen has been studied as part of maintenance protocols following intensive chemotherapy or as part of lower-intensity regimens for older adults with Ph+ ALL[2][3][4][5].
