Dato + Tras-vc-MMAE is an experimental bispecific antibody-drug conjugate (bsADC) designed to simultaneously target Human Epidermal Growth Factor Receptor 2 (HER2) and Trophoblast Cell-Surface Antigen 2 (Trop-2). The construct utilizes a monovalent bispecific IgG1 framework incorporating the Fab fragments of datopotamab (Dato) and trastuzumab (Tras). It is conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a lysosomally cleavable valine-citrulline (vc) linker, with an average drug-to-antibody ratio (DAR) of 4.2. By targeting two distinct tumor-associated antigens, the agent aims to enhance binding affinity, improve internalization, and overcome resistance mechanisms associated with monospecific ADCs. Preclinical data indicates potent antitumor activity in models with high Trop-2 and varying HER2 expression levels, demonstrating improved efficacy compared to monotherapy ADCs targeting either antigen alone.
