Overview

This is a **combination regimen** composed of four agents: - **Datopotamab deruxtecan**: an antibody-drug conjugate (ADC) targeting *trophoblast cell-surface antigen 2* (TROP2) with a topoisomerase I inhibitor payload (deruxtecan). It selectively binds TROP2 on cancer cells, is internalized, and releases its cytotoxic payload, causing DNA damage and cell death. Datopotamab deruxtecan has shown activity in multiple tumor types, notably breast and non-small cell lung cancer. - **5-fluorouracil (5-FU)**: a pyrimidine analog antimetabolite chemotherapy that inhibits thymidylate synthase and thereby DNA synthesis, leading to cytotoxicity in rapidly dividing cells. - **Leucovorin**: a reduced folate used to enhance the cytotoxic effects of 5-FU by stabilizing its binding to thymidylate synthase. - **Bevacizumab**: a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and limiting tumor blood supply. This combination is investigational; it includes an approved ADC with standard chemotherapeutics and an anti-angiogenic monoclonal antibody, aimed at maximizing efficacy through synergistic targeting of multiple cancer cell vulnerabilities.
