Overview

This is a **combination regimen of datopotamab deruxtecan, carboplatin, and bevacizumab**, used in oncology clinical trials for the treatment of advanced solid tumors, including non-small cell lung cancer and ovarian cancer. - **Datopotamab deruxtecan** is an antibody-drug conjugate composed of a monoclonal antibody targeting TROP2 (trophoblast cell-surface antigen-2/TACSTD2), conjugated via a tetrapeptide-based cleavable linker to a topoisomerase I inhibitor payload (DXd). It induces selective tumor cell death, immunogenic cell death, and bystander cytotoxicity by binding to TROP2 on tumor cells, internalization, and intracellular release of DXd via lysosomal proteases, resulting in DNA damage and apoptosis of TROP2-expressing cells[2][5][7][8]. - **Carboplatin** is a small molecule platinum-based alkylating agent that induces DNA crosslinking and apoptosis by binding to guanine bases in DNA, interfering with DNA replication and transcription. - **Bevacizumab** is a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis essential for tumor growth and survival[3]. This combination is being clinically evaluated to achieve synergistic efficacy by integrating targeted cytotoxicity, DNA-damaging chemotherapy, and antiangiogenic activity.
