This combination consists of **datopotamab deruxtecan**, a Trop-2-directed antibody–drug conjugate (ADC) utilizing a monoclonal antibody targeting Trop-2 and delivering a topoisomerase I inhibitor payload, and **sabestomig**, a bispecific monoclonal antibody targeting **PD-1** (programmed cell death protein 1) and **TIM-3** (T cell immunoglobulin and mucin domain-containing protein 3)[1][2][3][4][5][6][7][8]. Datopotamab deruxtecan binds Trop-2 on cancer cells, internalizes, and releases deruxtecan to block topoisomerase I, causing DNA damage and cell death[1][3][5][7]. Sabestomig blocks PD-1 and TIM-3, checkpoint proteins that inhibit T cell activation, thereby enhancing the immune system's anti-tumor activity[2][4][6][8]. Both are investigational and/or approved therapies for various advanced solid tumors, breast cancer, and Hodgkin lymphoma. No clinical trial or regulatory record exists for their combined use as a combination regimen, but components are both in active late-stage cancer development.
