Overview
This investigational combination consists of three agents: - **Datopotamab deruxtecan (Dato-DXd)**, an antibody-drug conjugate targeting Trop-2 protein, delivers the cytotoxic payload deruxtecan (a DNA topoisomerase I inhibitor) via a protease-cleavable linker, leading to DNA damage and apoptosis in Trop-2-expressing tumor cells.[2][4][5] - **Volrustomig (MEDI5752)**, is a bispecific monoclonal antibody targeting PD-1 and CTLA-4, designed to enhance antitumor immune responses (PD-1/CTLA-4 checkpoint inhibition). - **Carboplatin**, a platinum-containing compound, causes DNA crosslinking and inhibits DNA synthesis, functioning as a cytotoxic chemotherapy agent.[3] This triple combination is investigated for the treatment of non-small cell lung cancer (NSCLC) in Phase 1 clinical trials, aiming to leverage targeted cytotoxicity, immune checkpoint blockade, and platinum-based chemotherapy.[1]
