This combination chemotherapy regimen, consisting of daunorubicin and cytarabine, was evaluated by Assistance Publique - Hôpitaux de Paris (AP-HP) in a Phase 3 clinical trial specifically for patients with Core Binding Factor Acute Myeloid Leukemia (CBF-AML). Daunorubicin is an anthracycline antineoplastic antibiotic that exerts its effects through DNA intercalation and the inhibition of topoisomerase II, leading to DNA strand breaks. Cytarabine is a pyrimidine nucleoside analog that, upon phosphorylation to cytarabine triphosphate, competes with deoxycytidine triphosphate for incorporation into DNA, thereby inhibiting DNA polymerase and causing premature chain termination. The AP-HP study (NCT00428558) investigated a timed-sequential induction (TSI) schedule to optimize the synergistic effects of these agents in adult patients with t(8;21) or inv(16) chromosomal translocations.
