Drug intelligence / Profile preview
Vyxeos (daunorubicin + cytarabine) is a liposomal combination chemotherapy developed by Jazz Pharmaceuticals for the treatment of specific types of acute myeloid leukemia (AML). It consists of two established antineoplastic agents, daunorubicin and cytarabine, encapsulated within a liposome at a fixed 1:5 molar ratio. This specific ratio has been shown in preclinical studies to maximize synergistic cell kill. The liposomal formulation, developed using proprietary CombiPlex technology, is designed to deliver the drug combination directly to the bone marrow, where it is preferentially taken up by leukemia cells, thereby increasing efficacy and potentially reducing systemic toxicity. Daunorubicin is an anthracycline that inhibits topoisomerase II and intercalates into DNA, while cytarabine is a nucleoside analog that inhibits DNA polymerase and interferes with DNA synthesis. Vyxeos is primarily indicated for newly-diagnosed therapy-related AML (t-AML) or AML with myelodysplasia-related changes (AML-MRC).
