Drug intelligence / Profile preview
daunorubicin + cytarabine liposome (JZP351, CPX-351, Vyxeos) is a dual-drug liposomal formulation encapsulating daunorubicin and cytarabine at a fixed 1:5 molar ratio, designed to maintain synergistic drug exposure and improve delivery to leukemia cells. This formulation is approved for the treatment of newly diagnosed therapy-related acute myeloid leukemia (AML) or AML with myelodysplasia-related changes. The liposomal encapsulation enhances preferential delivery to malignant cells, provides sustained drug exposure, and is associated with improved remission rates and overall survival compared to conventional cytarabine plus daunorubicin ("7+3") regimens in high-risk AML populations. Key features include reduced gastrointestinal and cardiac toxicities compared to conventional formulations. The mechanism involves inhibition of DNA synthesis and function in leukemia cells through the combined actions of a nucleoside analog (cytarabine) and a topoisomerase II inhibitor (daunorubicin)[1][2][3][7].
