Drug intelligence / Profile preview
Daunorubicin and cytarabine liposome is a synergistic fixed-ratio liposomal formulation of the anthracycline daunorubicin and the antimetabolite cytarabine. Encapsulated in a 1:5 molar ratio, this formulation is designed to optimize the delivery of these two cytotoxic agents to bone marrow blasts while minimizing systemic exposure and toxicity. Daunorubicin acts by intercalating DNA and inhibiting topoisomerase II, while cytarabine is a pyrimidine analog that inhibits DNA polymerase and DNA synthesis. The liposomal delivery system extends the plasma half-life of the drugs and facilitates their preferential uptake by leukemic cells. It is primarily indicated for the treatment of adults with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). CSPC Zhongnuo Pharmaceutical is developing a version of this liposomal combination in China, currently in Phase 3 clinical trials for high-risk secondary AML.
