Drug intelligence / Profile preview
DaunoXome is a liposomal formulation of daunorubicin citrate, an anthracycline chemotherapy agent. Developed by NeXstar Pharmaceuticals, it was specifically designed to improve the therapeutic index of daunorubicin by utilizing a liposomal delivery system. This nanoparticle modality enhances drug delivery to tumor tissues, such as those in Kaposi's sarcoma, by exploiting the increased permeability of tumor vasculature while simultaneously reducing systemic toxicities, particularly cardiotoxicity, compared to conventional daunorubicin. The drug exerts its antineoplastic effect by intercalating between DNA base pairs and inhibiting topoisomerase II, thereby disrupting DNA replication and RNA synthesis. It is primarily indicated as a first-line cytotoxic therapy for advanced, HIV-associated Kaposi's sarcoma.
