DB-1303 + pertuzumab is an experimental combination therapy involving two agents: **DB-1303**, a next-generation HER2-targeting antibody-drug conjugate (ADC), and **pertuzumab**, a monoclonal antibody targeting HER2. DB-1303 is composed of a humanized anti-HER2 IgG1 monoclonal antibody linked to a proprietary DNA topoisomerase I inhibitor via a cleavable linker, with a high drug-to-antibody ratio. Pertuzumab is a HER2 dimerization inhibitor that binds to a distinct epitope on the HER2 extracellular domain, preventing receptor dimerization. The combination aims to enhance antitumor activity in **HER2-expressing cancers** by dual blockade of HER2 signaling and delivering a cytotoxic payload directly to cancer cells. DB-1303 is being developed by DualityBio (with BioNTech as a collaborator), with pertuzumab developed and manufactured by Roche. The combination is being studied primarily for breast cancer, especially HER2-low and HER2-positive subtypes, and may be explored in other HER2-expressing solid tumors[1][2][4][6].
