DB-1310 + trastuzumab is an investigational combination therapy consisting of DB-1310, a HER3-targeting antibody-drug conjugate (ADC), and trastuzumab, a monoclonal antibody targeting HER2. Preclinical and early clinical studies demonstrate that this combination synergistically inhibits the proliferation of breast cancer cells with high HER2 and HER3 expression by enhancing the internalization of DB-1310, disrupting HER2/HER3 dimerization, and increasing payload delivery into tumor tissue. DB-1310 exerts cytotoxicity upon internalization and payload release, while trastuzumab enhances efficacy by augmenting DB-1310 uptake and blocking HER2-mediated signaling. This strategy potentially overcomes resistance observed with trastuzumab alone and provides robust tumor suppression in HER2+/HER3+ cancers. Developed primarily for breast cancer but also in study for other advanced/metastatic solid tumors[1][3][5].
