DEB-TACE + sorafenib is a combination therapy used primarily for the treatment of unresectable advanced hepatocellular carcinoma (HCC). It combines drug-eluting bead transarterial chemoembolization (DEB-TACE), a locoregional therapy that delivers chemotherapy directly to liver tumors via embolized beads loaded with doxorubicin, with systemic administration of sorafenib, an oral multikinase inhibitor. Sorafenib inhibits tumor cell proliferation and angiogenesis by targeting multiple kinases including RAF kinase, vascular endothelial growth factor receptors (VEGFR), and platelet-derived growth factor receptor (PDGFR). The combination aims to enhance antitumor efficacy by combining local tumor ischemia and chemotherapy from DEB-TACE with systemic inhibition of tumor growth pathways by sorafenib. Clinical studies have shown this combination to be generally well tolerated with manageable toxicities mostly related to sorafenib, demonstrating promising disease control rates in patients with advanced HCC[1][2][3].
