Drug intelligence / Profile preview
Debio-0512 is a long-acting, 6-month sustained-release formulation of triptorelin pamoate, a synthetic decapeptide analog of gonadotropin-releasing hormone (GnRH). Developed by Debiopharm, this formulation utilizes poly-lactic-co-glycolic acid (PLGA) microsphere technology to provide a continuous release of the active ingredient over a 24-week period. The drug's mechanism of action involves the potent stimulation of GnRH receptors in the pituitary gland, which initially increases the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). However, chronic exposure leads to the down-regulation and desensitization of these receptors, resulting in a profound suppression of LH and FSH. This suppression subsequently reduces serum testosterone to castrate levels (typically <50 ng/dL), thereby inhibiting the growth of hormone-sensitive prostate cancer cells. Debio-0512 is primarily indicated for the palliative treatment of advanced prostate cancer and offers the clinical benefit of less frequent dosing compared to 1-month or 3-month formulations.
