Decitabine + anti-PD-1 is a combination therapy consisting of the DNA hypomethylating agent decitabine and an anti–programmed death 1 (PD-1) monoclonal antibody (such as camrelizumab or sintilimab). Decitabine acts as an epigenetic modulator by inhibiting DNA methyltransferase, leading to hypomethylation of DNA and reactivation of silenced genes involved in immune response. Anti-PD-1 antibodies block the PD-1 receptor on T cells, preventing tumor-induced T cell exhaustion and restoring antitumor immunity. The combination has shown synergistic effects in preclinical models and clinical studies by enhancing T cell function, increasing tumor-infiltrating lymphocytes, reversing resistance to PD-1 blockade, and improving response rates in relapsed/refractory classical Hodgkin lymphoma (cHL), myelodysplastic syndromes (MDS), non-small cell lung cancer (NSCLC), and other malignancies[1][2][3][4][5][6][7].
