Overview

This is an **experimental combination therapy** consisting of three chemotherapeutic agents: **decitabine**, **gemcitabine**, and **oxaliplatin**. Decitabine is a pyrimidine nucleoside analogue that inhibits DNA methyltransferase, resulting in DNA hypomethylation and altered gene expression, ultimately interfering with tumor cell proliferation. Gemcitabine is a nucleoside metabolic inhibitor that incorporates into DNA, causing chain termination and apoptosis. Oxaliplatin is a platinum-based agent that forms DNA adducts, inhibiting DNA replication and transcription and inducing cell death. The combination is being investigated for enhanced antitumor activity via synergistic effects on DNA synthesis, methylation, and repair mechanisms. Preclinical and early clinical work suggest potential benefit in hematologic malignancies and solid tumors, but this triple-drug regimen is not yet standard of care.[3][9][10]
