Overview

Decitabine + homoharringtonine + cytarabine (DHC) is an investigational combination chemotherapy regimen primarily developed and studied by researchers at The First Affiliated Hospital of Chongqing Medical University for the treatment of acute myeloid leukemia (AML). This regimen integrates three distinct therapeutic mechanisms to achieve synergistic antileukemic effects: decitabine, a hypomethylating agent, inhibits DNA methyltransferase (DNMT) to reactivate silenced tumor suppressor genes; homoharringtonine (also known as omacetaxine mepesuccinate) is a plant alkaloid that inhibits protein translation by binding to the ribosomal A-site; and cytarabine is an antimetabolite that inhibits DNA polymerase and disrupts DNA synthesis. The combination is particularly focused on improving outcomes in elderly patients or those with newly diagnosed AML who may not tolerate intensive standard induction therapy.
