Decitabine + toripalimab is a combination therapy consisting of the hypomethylating agent decitabine and the anti-PD-1 monoclonal antibody toripalimab. Decitabine is a DNA methyltransferase inhibitor that induces the re-expression of silenced genes, including those involved in antigen presentation and immune recognition. This epigenetic modification is hypothesized to sensitize tumor cells to immune checkpoint inhibition. Toripalimab is a humanized monoclonal antibody that targets the Programmed Cell Death Protein 1 (PD-1) receptor on T cells, blocking its interaction with ligands PD-L1 and PD-L2 to restore anti-tumor immune responses. The combination is primarily being investigated for the treatment of hematological malignancies, such as recurrent or refractory follicular lymphoma, where epigenetic priming may overcome resistance to immunotherapy.
