Deferasirox + calcium-vitamin d3 is a non-standard combination of an iron chelator (deferasirox) and a dietary supplement containing calcium and vitamin D3 (cholecalciferol). Deferasirox is an orally active tridentate ligand that selectively binds ferric iron (Fe3+), forming a stable complex that is excreted primarily via the feces, thereby reducing chronic iron overload in patients receiving frequent blood transfusions or with non-transfusion-dependent thalassemia syndromes. Calcium-vitamin D3 supplements are commonly used to support bone health, with calcium serving as a mineral essential for bone structure and vitamin D3 facilitating intestinal absorption of calcium. There is some evidence suggesting potential synergy between deferasirox and vitamin D in promoting monocyte differentiation in acute myeloid leukemia cells, but this combination is not standard therapy. Coadministration may present drug interaction risks; specifically, concurrent use of oral deferasirox with antacids or supplements containing polyvalent cations like calcium can reduce the absorption and efficacy of deferasirox.
