A combination iron chelation therapy consisting of two oral iron chelators, deferasirox and deferiprone. This combination is used primarily for treating transfusion-induced iron overload in patients with conditions like β-thalassemia major, particularly those with suboptimal response to monotherapy. Deferasirox is a tridentate iron chelator that binds iron in a 2:1 ratio and is excreted primarily through feces, while deferiprone is a bidentate chelator requiring three molecules to bind one iron ion and is excreted through urine. The combination has demonstrated superior efficacy in reducing serum ferritin levels and improving cardiac iron removal compared to monotherapy in several clinical studies.
