Overview

deferasirox + rifampicin is a combination of two drugs: deferasirox, an oral iron chelator primarily used to treat chronic iron overload due to blood transfusions, and rifampicin (also known as rifampin), a potent antibiotic and UDP-glucuronosyltransferase (UGT) enzyme inducer most commonly used to treat tuberculosis and other bacterial infections. This combination lacks an approved or recognized fixed-dose formulation but may be encountered when both drugs are co-administered. Concomitant use is generally not recommended due to significant drug-drug interactions whereby rifampicin (a potent UGT inducer) markedly reduces the exposure and efficacy of deferasirox by inducing its metabolic clearance, resulting in a decrease in deferasirox systemic exposure by approximately 44%. The clinical reason for using this combination is typically for distinct indications rather than for a synergistic pharmacological effect. Close monitoring of iron levels and possible dose adjustment of deferasirox are required if in rare circumstances this combination cannot be avoided.
