Overview

This combination therapy regimen consists of the iron-chelating agent deferoxamine (DFO) administered in conjunction with conventional transarterial chemoembolization (TACE). It is primarily investigated for the treatment of unresectable hepatocellular carcinoma (HCC). Deferoxamine acts by binding to free iron, thereby depleting the intracellular iron stores necessary for DNA synthesis and cell proliferation in rapidly dividing tumor cells. TACE is a localized interventional procedure that involves the delivery of cytotoxic chemotherapy (typically doxorubicin or cisplatin) directly into the hepatic artery supplying the tumor, followed by the administration of embolic agents to induce ischemia and tumor necrosis. The rationale for this combination is that iron depletion via deferoxamine may sensitize HCC cells to the cytotoxic and ischemic effects of TACE, potentially improving therapeutic outcomes in patients with advanced liver cancer.
