Overview

DEI-LT (pPJV2012) is an investigational DNA vaccine candidate developed for the prevention of diarrhea caused by enterotoxigenic *Escherichia coli* (ETEC). The vaccine utilizes the pPJV2012 plasmid, which encodes both the A and B subunits of the ETEC heat-labile enterotoxin (LT). It is designed for delivery via a biolistic "gene gun" device, which uses compressed helium to propel DNA-coated gold microparticles into the epidermal layer of the skin. Upon entry into host cells, the plasmid DNA is transcribed and translated into the LT protein, which serves as an antigen to stimulate the production of toxin-neutralizing antibodies. This mechanism aims to neutralize the LT toxin before it can bind to intestinal epithelial cells and trigger the signaling cascades that lead to fluid secretion and diarrhea. Clinical development has primarily been led by the U.S. Naval Medical Research Center in collaboration with technology partners.
