Delavirdine + didanosine is a combination of two antiretroviral drugs used in the treatment of human immunodeficiency virus type 1 (HIV-1) infection. Delavirdine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that binds directly to the HIV-1 reverse transcriptase enzyme, blocking its polymerase activities. Didanosine is a nucleoside reverse transcriptase inhibitor (NRTI) that is phosphorylated intracellularly to its active metabolite, dideoxyadenosine triphosphate (ddATP), which then inhibits reverse transcriptase by competing with natural substrates and terminating viral DNA chain elongation. This combination was studied for its synergistic activity against HIV-1, with clinical trials demonstrating enhanced antiretroviral efficacy compared to monotherapy.
