Overview

Delolimogene mupadenorepvec + atezolizumab is an experimental combination immunotherapy regimen. - Delolimogene mupadenorepvec (LOAd703) is an oncolytic adenovirus gene therapy genetically engineered to express membrane-bound CD40 ligand and full-length 4-1BB ligand (4-1BBL), which potently stimulate anti-tumor immune responses. It is delivered by intratumoral injection and is intended to act as an immunostimulatory and oncolytic agent to generate a local and systemic immune response against tumors[2][4]. - Atezolizumab (Tecentriq) is a humanized monoclonal antibody that targets Programmed cell death ligand 1 (PD-L1), acting as an immune checkpoint inhibitor to enhance T cell-mediated anti-tumor activity. The combination aims to synergistically enhance both innate and adaptive anti-tumor immunity, with delolimogene mupadenorepvec initiating immune responses within the tumor and atezolizumab releasing immune suppression in the tumor microenvironment. Active development includes Phase I/II trials in multiple solid tumors (including pancreatic, biliary, colorectal, and ovarian cancers), often in combination with standard chemotherapy.
