Delta3,2-hydroxybakuchiol is a bakuchiol analog isolated from Fructus Psoraleae. It functions as a potent and efficacious monoamine transporter inhibitor, primarily targeting the dopamine transporter (DAT) and norepinephrine transporter (NET) with high selectivity over the serotonin transporter (SERT). It competitively inhibits DAT and NET with partial reversibility and occupies cocaine binding sites. Preclinical studies have shown its ability to counteract 1-methyl-4-phenylpyridinium-induced toxicity in cells expressing DAT, increase activity in intact mice, and improve decreased activity in reserpinized mice, suggesting potential therapeutic applications for disorders such as Parkinson's disease, depression, and cocaine addiction.
