Demcizumab + paclitaxel is a **combination therapy** involving demcizumab, a humanized monoclonal antibody that targets Delta-like ligand 4 (DLL4), and paclitaxel, a widely used chemotherapeutic agent. **Demcizumab** is an IgG2 monoclonal antibody that specifically inhibits DLL4, thereby blocking Notch signaling, which is implicated in cancer stem cell maintenance, tumor growth, and angiogenesis. This blockade leads to reduced cancer stem cell frequency, inhibition of tumor growth, and antiangiogenic effects. When combined with **paclitaxel**, which stabilizes microtubules and inhibits mitosis, preclinical and early clinical data show evidence of synergistic antitumor activity and manageable toxicity. The combination is being studied primarily in patients with platinum-resistant epithelial ovarian cancer, as well as in other tumor types such as pancreatic cancer.
