An investigational **autologous dendritic-cell cancer vaccine** prepared by loading dendritic cells with messenger RNA encoding human telomerase reverse transcriptase and survivin, together with tumor-cell-derived mRNA. The transfected dendritic cells are intended to present these tumor-associated antigens and stimulate antigen-specific antitumor T-cell responses against melanoma cells expressing hTERT, survivin, and patient-derived tumor antigens. It was evaluated in an investigator-sponsored Phase I/II study in metastatic malignant melanoma; the registered study was terminated. The broader protocol also used temozolomide before vaccination and offered lymphodepletion followed by ex vivo-expanded autologous T-cell reinfusion to patients demonstrating an immune response.
