Denenicokin + nivolumab is an investigational combination therapy consisting of **denenicokin**, an engineered cytokine, and **nivolumab**, a monoclonal antibody targeting programmed death-1 (PD-1). **Nivolumab** is a **PD-1 immune checkpoint inhibitor** that blocks the PD-1 pathway, thereby enhancing T-cell-mediated anti-tumor immune responses. **Denenicokin** is an engineered interleukin-18 (IL-18) variant (also known as WTX-124) designed to activate the interleukin-18 receptor pathway, stimulating an immune response against cancer cells even in the presence of natural inhibitors such as IL-18 binding protein. The combination aims to produce synergistic immunotherapeutic effects by activating and unleashing cytotoxic T-cell and natural killer cell responses, potentially enhancing anti-cancer immune activity in patients refractory to checkpoint inhibition.
