Dengvaxia (CYD-TDV) is a live-attenuated tetravalent vaccine developed by Sanofi Pasteur for the prevention of dengue fever caused by dengue virus serotypes 1, 2, 3, and 4. It is constructed using the ChimeriVax technology, which utilizes the yellow fever 17D vaccine virus as a genetic backbone. The genes encoding the pre-membrane (prM) and envelope (E) proteins of the yellow fever virus are replaced with those from the four dengue virus serotypes. This chimeric approach allows the vaccine to induce neutralizing antibodies against all four dengue serotypes while maintaining the attenuation profile of the yellow fever 17D strain. It is primarily indicated for individuals aged 6 to 45 years (depending on the region) living in endemic areas who have had a laboratory-confirmed previous dengue infection, as it can increase the risk of severe dengue in seronegative individuals.
