Drug intelligence / Profile preview
Denikitug + nivolumab is an investigational combination immunotherapy regimen consisting of denikitug (GS-1811), an afucosylated humanized monoclonal antibody targeting C-C chemokine receptor type 8 (CCR8), and nivolumab (Opdivo), an approved anti-programmed cell death protein 1 (PD-1) monoclonal antibody. Denikitug is designed to selectively deplete immunosuppressive regulatory T cells (Tregs) in the tumor microenvironment via antibody-dependent cellular cytotoxicity (ADCC). Nivolumab blocks the PD-1/PD-L1 checkpoint pathway to restore T-cell-mediated anti-tumor immune responses. Developed by Gilead Sciences, this combination is being evaluated in Phase 2 clinical trials for the treatment of advanced solid tumors, including HER2-negative, unresectable or metastatic gastric, gastroesophageal junction (GEJ), and esophageal adenocarcinomas, as well as advanced microsatellite-stable (MSS) colorectal cancer.
