denosumab + nivolumab

Denosumab + nivolumab is an investigational combination therapy consisting of two monoclonal antibodies: denosumab, a fully human IgG2 antibody targeting RANK ligand (RANKL), and nivolumab, a fully human IgG4 antibody targeting programmed cell death protein 1 (PD-1). Denosumab inhibits osteoclast-mediated bone resorption by blocking the interaction between RANKL and its receptor RANK, thereby reducing bone loss and skeletal-related events in cancer patients with bone metastases. Nivolumab is an immune checkpoint inhibitor that blocks PD-1 on T cells, preventing tumor-induced immunosuppression and restoring anti-tumor immune responses. The combination is being studied for potential synergistic effects in enhancing anti-tumor immunity, particularly in cancers such as metastatic melanoma and non-small cell lung cancer (NSCLC) with bone metastases[2][4][6].
