Depatuxizumab mafodotin + temozolomide is a combination therapy investigated primarily for the treatment of glioblastoma, particularly in patients whose tumors exhibit amplification of the epidermal growth factor receptor (EGFR). Depatuxizumab mafodotin (Depatux-M, formerly ABT-414) is an antibody-drug conjugate composed of a monoclonal antibody targeting activated EGFR linked to the cytotoxic agent monomethylauristatin F (MMAF), which disrupts microtubule function. Temozolomide is an oral alkylating agent that induces DNA damage and cell death. The combination has been studied in both newly diagnosed and recurrent glioblastoma, with clinical trials showing encouraging antitumor activity and manageable toxicity profiles. The most common adverse events are ocular toxicities related to the MMAF payload and hematologic effects from temozolomide. Despite promising results in phase II studies, further development of this combination has been discontinued by its developer.
