Drug intelligence / Profile preview
DepoCyte is a sustained-release liposomal formulation of the antimetabolite cytarabine (Ara-C), specifically engineered for intrathecal administration into the cerebrospinal fluid. Utilizing the proprietary DepoFoam multivesicular liposome technology, it provides a slow release of cytarabine, maintaining cytotoxic concentrations in the neuroaxis for up to 14 days, which significantly reduces the frequency of injections compared to standard cytarabine. The active ingredient, cytarabine, is a pyrimidine analog that, upon intracellular phosphorylation, inhibits DNA polymerase and incorporates into DNA, leading to S-phase specific cell death. While it received regulatory approval for the treatment of lymphomatous meningitis, it has been investigated in clinical trials such as the GMALL Elderly 1/2003 study led by Goethe University for central nervous system prophylaxis and treatment in elderly patients with acute lymphoblastic leukemia (ALL). However, the product was discontinued globally in 2017 due to persistent technical manufacturing complexities.
