This combination therapy is used in clinical research settings to study the effects of hormonal suppression and replacement in women with Polycystic Ovary Syndrome (PCOS). It consists of depot leuprolide, a gonadotropin-releasing hormone (GnRH) agonist that temporarily suppresses the hypothalamic-pituitary-ovarian axis to reduce endogenous production of estrogen and testosterone. To prevent symptoms of hormone deficiency and maintain physiological female hormone levels, it is co-administered with exogenous estrogen and progestin (typically via a transdermal patch). This "add-back" regimen allows researchers to isolate the effects of hyperandrogenism or other PCOS-related factors while maintaining a controlled hormonal environment.
