Overview

Desipramine + naltrexone is a combination of two small molecule drugs, each with distinct mechanisms of action. Desipramine is a tricyclic antidepressant primarily used for the treatment of depression. It acts mainly as a potent inhibitor of norepinephrine reuptake and, to a lesser extent, serotonin reuptake in the central nervous system. This leads to increased levels of these neurotransmitters in the synaptic cleft and an overall enhancement in serotonergic neurotransmission. Desipramine also has antagonistic effects on histamine-H1 receptors, alpha-1 adrenergic receptors, and muscarinic receptors[1][4]. Naltrexone is an opioid receptor antagonist used primarily for treating alcohol use disorder and opioid use disorder by blocking the rewarding effects of opioids and alcohol at opioid receptors throughout the brain[6]. The combination does not have an established unique indication or brand name; it may be considered experimentally or off-label for research into interactions between antidepressants and opioid systems[2].
