Overview

This drug is a combination of desloratadine, a second-generation H1-receptor antagonist, and ranitidine, an H2-receptor antagonist. It was evaluated by Hamilton Health Sciences Corporation as a premedication strategy for children aged 5-10 years with peanut allergy undergoing oral immunotherapy (OIT) in the PISCES trial (NCT01601522). The combination was intended to reduce the frequency and severity of allergic side effects, specifically skin and gastrointestinal reactions, that often occur during the dose-escalation phase of desensitization. Desloratadine provides long-acting, non-sedating blockade of H1 receptors to mitigate systemic allergic symptoms, while ranitidine blocks H2 receptors to address gastrointestinal distress and gastric acid secretion associated with the ingestion of allergen proteins.
