This is a **combination regimen** consisting of devimistat (CPI-613), modified FOLFIRINOX, and bevacizumab. - **Devimistat** is a novel lipoate analog targeting tumor mitochondrial metabolism, acting as an inhibitor of key mitochondrial enzymes involved in energy production, notably pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase complexes. - **Modified FOLFIRINOX** is a chemotherapy regimen consisting of lower or adjusted doses of oxaliplatin, irinotecan, leucovorin (folinic acid), and fluorouracil (5-FU) compared to the original FOLFIRINOX protocol, aimed at improving tolerability while maintaining efficacy; it is widely used as first-line treatment for metastatic pancreatic cancer and other GI malignancies[4][6]. - **Bevacizumab** is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thus blocking angiogenesis in tumors. This three-drug combination is under investigation for advanced/metastatic pancreatic cancers and potentially for other GI malignancies.
