Drug intelligence / Profile preview
Dexamethasone is a potent synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties. In the context of Sun Pharmaceutical Industries (following its acquisition of InSite Vision), it was formulated as a 0.1% ophthalmic suspension utilizing the proprietary DuraSite drug delivery vehicle. DuraSite is a mucoadhesive polymer-based system designed to extend the residence time of the drug on the ocular surface, thereby improving bioavailability and potentially allowing for less frequent dosing. This specific formulation, known as ISV-205, was evaluated in Phase 3 clinical trials for the treatment of blepharoconjunctivitis, both as a standalone therapy and as a comparator arm for the combination product AzaSite Plus (azithromycin/dexamethasone). Dexamethasone exerts its effect by binding to the glucocorticoid receptor, leading to the modulation of gene expression that inhibits the production of pro-inflammatory cytokines and enzymes.
