Drug intelligence / Profile preview
Dexamethasone + buprenorphine is a combination of two pharmaceutical agents used as adjuvants in regional anesthesia, particularly for prolonging the duration and improving the quality of nerve blocks. Dexamethasone is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressant properties, acting primarily by modulating gene expression to suppress pro-inflammatory cytokines. Buprenorphine is a semi-synthetic opioid that acts as a partial agonist at the mu-opioid receptor and an antagonist at the kappa-opioid receptor, providing analgesia with reduced risk of respiratory depression compared to full opioid agonists. When administered perineurally (around nerves) together with local anesthetics such as bupivacaine, this combination has been shown to prolong block duration, reduce pain scores, and decrease postoperative opioid consumption without increasing adverse effects compared to intravenous administration[1][3][7]. The combination does not have an established brand name or widespread regulatory approval for use together but has been studied in clinical trials for perioperative pain management.
