Overview

**dexamethasone + melphalan** is a combination chemotherapy regimen commonly used for treating multiple plasma cell disorders, most notably **multiple myeloma**, **primary systemic (AL) amyloidosis**, and **POEMS syndrome**. **Dexamethasone** is a synthetic glucocorticoid corticosteroid that suppresses inflammation and immune responses, and **melphalan** is a bifunctional alkylating agent that induces cytotoxic DNA damage and cell death in rapidly dividing cells. Together, these drugs act synergistically to inhibit clonal plasma cell proliferation, reduce tumor burden, and ameliorate disease symptoms. Clinical studies have demonstrated significant hematologic and neurologic responses with this combination in plasma cell dyscrasias, with manageable toxicity[1][2][5].
